In non-stressed rats, Tianeptine Sodium treatment prevented the increase in CMS-induced adrenocorticotropin-releasing factor (CRF) mRNA levels in dBNST and reduced the CRF mRNA levels in dBNST. Tianeptine treatment significantly reduced the CRF mRNA levels of ventral BNST and CEA in the pressure-avoidance control group and CMS exposed rats. Tianeptine blocked stress-induced PB enhancement in CA1 region in the basolateral nucleus of the amygdala (BLA) of anesthetized rats, but did not affect stress-induced enhancement of LTP. Tianeptine was administered with enhanced PB enhancement under non-stress conditions in the amygdala, hippocampus, and LTP of anesthetized rats. In rats, Tianeptine attenuates a behavioral sign of peripheral induction, but not central, LPS or IL-1β -induced pathological behavior. In rats, Tianeptine causes NMDA receptors to normalize the amplitude ratio of AMPA/ red alginate receptor-mediated currents and prevents stress-induced attenuation of NMDA-EPscs inactivation. Tianeptine significantly reduced apoptosis in the temporal cortex and dentate gyrus of the hippocampus in both control and stressed animals, but had no effect on Horn of Africa Ammons. Tianeptine (2.5 mg/kg intraperitoneal injection) increases extracellular dopamine only in the nucleus accumbens. Tianeptine significantly increased extracellular concentrations of hydroxyphenylacetic acid (DOPAC) and hypervanilic acid (HVA) in both nucleus accumbens and striatum.
